What NAD⁺ Actually Is — and Why It Comes Up in Every Conversation About Aging
If you have read anything about longevity in the last few years, you have run into three letters over and over: NAD⁺. It shows up in research summaries, in podcasts, and on an increasing number of supplement labels — usually without anyone stopping to explain what it is.
Here is the plain version.
NAD⁺ is a coenzyme your cells cannot work without
NAD⁺ — nicotinamide adenine dinucleotide — is a coenzyme found in every living cell. Its job is to carry electrons from one reaction to another, which is the mechanism your mitochondria use to turn food into usable energy. It also acts as a substrate for a family of enzymes involved in normal cellular maintenance.
Two things make it interesting to researchers who study aging.
The first is that NAD⁺ is not optional. Cells cannot produce energy through normal metabolic pathways without it.
The second is that measured NAD⁺ levels in human tissue tend to be lower in older adults than in younger ones. That observation is what started the current wave of research, and it is the honest reason NAD⁺ is discussed in the context of aging — not because anyone has shown that topping it up rewinds a clock.
What the research does and does not establish
This is the part most articles skip, so let us be direct.
Human trials on NAD⁺ precursors have generally looked at whether supplementation raises NAD⁺ levels in blood or tissue. Several have found that it does. That is a meaningful finding, and it is also a narrower finding than the headlines suggest.
What has not been established is that raising NAD⁺ levels produces a specific health outcome in humans — more years, fewer illnesses, reversed aging. Anyone telling you otherwise is ahead of the evidence.

Why precursors, rather than NAD⁺ itself
NAD⁺ is a large molecule and is not well absorbed intact. So supplements generally supply a precursor — a smaller compound the body converts into NAD⁺ through its own pathways.
Nicotinamide riboside is one of the most studied. It is a form of vitamin B3 that enters the NAD⁺ salvage pathway, and it is the precursor with the largest body of published human data behind it.
You may also see nicotinamide mononucleotide (NMN), which is a step further along the same pathway, and plain niacin or niacinamide, which are older and cheaper forms of B3 that also feed into NAD⁺ production.
What to look for on a label
If you are comparing products, four things are worth checking:
- Which precursor, named specifically. "NAD⁺ complex" tells you nothing.
Nicotinamide riboside chloride, NMN, or niacinamide tells you something.
- Whether it is disclosed at all. Many formulas bury the precursor inside a
proprietary blend where individual amounts are not stated.
- What else is in the formula. NAD⁺ pathways do not run in isolation — they
depend on B vitamins the body also needs from other sources.
- Whether the claims are careful. A label that promises to reverse aging is
telling you something about the company, not the ingredient.
